In Europe, the batch release process outlined in EudraLex Volume 4, Annex 16<\/a> appears simple enough: <\/p>\n But during the discussion, the QP revealed a typical challenge of legacy batch release processes<\/a>: separate sites for drug products, packaging, and API manufacturing interact during the batch production journey, making the final release process \u201ctedious\u201d and \u201ccumbersome\u201d when executed manually on paper.<\/p>\n Reviewing paper documents from multiple CMOs, combining them, and piecing together the batch genealogy step by step continues to demand high levels of time and effort from QPs and disposition owners. And because paper batch certificates and registers remain prevalent, the process has to be completed on-site using wet signatures and stamps.<\/p>\n As the organization looked to replace its legacy processes with modernized and digital batch release, it met several hurdles.<\/p>\n BioPhorum\u2019s 2026 Batch Disposition Manifesto<\/a> shows that there is industry-wide appetite for digital transformation, but for now regulators simply haven\u2019t kept pace.<\/p>\n The EMA released a four-page Q&A consultation document<\/a> about \u201cremote\u201d batch certification back in 2022, but there are still no formal EU guidelines for digital batch certification generally.<\/p>\n Whether digital batch certification is allowed at all depends on an organization\u2019s national competent authority, so European biopharmas aiming to digitize batch release may struggle to know where to start. <\/p>\n The picture is similar in the U.S. In the absence of a standalone remote batch release guidance document from the FDA, batch disposition guidance comes solely from the electronic records and signatures requirements of 21 CFR Part 11, and the quality control unit framework in 21 CFR Part 211. <\/p>\n For a fully digital process, the entire batch lifecycle should be available in a remotely accessible digital format. However, it takes concerted effort to digitize discrete sets of analog batch data from multiple contract manufacturing organizations (CMOs) and sites.<\/p>\n And full batch release modernization isn\u2019t just about converting paper to electronic records. To achieve the full efficiency benefits of optimized batch release<\/a>, quality leaders will need to systematically reimagine and reshape key processes \u2014 which means careful planning, gap analysis, and batch release solution scoping.<\/p>\n For these reasons, industry leaders are taking a phased approach to batch release transformation. The QP recommends three key actions.<\/p>\n In the absence of EU-wide regulation, the QP\u2019s organization made direct contact with its local competent GMP authority to discuss its batch release digitization plans, and secure awareness, alignment, and buy-in from the beginning of the project.<\/p>\n Dialog is actively continuing, with batch release process enhancements shared with the authority as they are developed for examination and feedback.<\/p>\n The QP makes a distinction between \u201cdigitization\u201d and \u201cdigitalization\u201d of batch release. These digitalization ingredients should form the primary targets of a batch release modernization project.<\/p>\n Although remote batch release with electronic batch registers and e-signatures is preferable to on-site paper processes, industry leaders view batch release transformation more holistically, and are moving even further to embed fully optimized and efficient batch processes.<\/p>\n For the QP and his organization, the final stage of full digital transformation is centralizing all batch activity in a purpose-built application.
